Multi-dimensional automated imaging detection analysis of cardiotoxic compounds

The diversity and complexity of cell detection methods are increasing in biological research and drug discovery. Stem cell-derived cells and tissues have become an increasingly attractive alternative to traditional in vitro and in vivo tests for drug development and toxicological safety assessment. In a study, human induced pluripotent stem cell (iPSC) -derived cardiomyocytes were used to develop functional and morphological readings to test the effects of different compounds in a multiparametric assay format.

The ImageXpress Pico Personal High Connotation Imaging Analysis System performs automated cell imaging and analysis of iPSC- derived cardiomyocytes to simultaneously determine calcium oscillation frequency, cell viability, cytoskeletal integrity, apoptosis, and mitochondrial function . The effect of calcium ion oscillation on the beating frequency of myocardial cells was studied by calcium ion oscillation measurement. A multi-parameter joint assessment of the results of different types of records provides a further understanding of the mechanisms of action of various compounds. These methods are characterized using a known group of cardiac active drugs and selected cardiotoxic compounds.
