Product introduction:
Enterobacteriaceae can reduce nitrate to nitrite. Nitrate reduction involves two processes: in the synthesis process, nitrate is reduced to nitrite and ammonia, and then ammonia is converted into amino acids and other nitrogenous compounds in cells. In the process of catabolism, nitrate or nitrite replaces oxygen as the terminal hydrogen acceptor in the respiratory enzyme system. The nitrate-reducing bacteria obtain oxygen from the nitrate to form nitrite and other reducing products.
The nitrate reduction experimental reagent is mainly composed of a p-aminobenzenesulfonic acid solution and an α-naphthylamine solution, and is mainly used for identifying Enterobacteria, Pseudomonas, and the like. Bacteria such as Escherichia coli can only reduce nitrate to nitrite, nitrite and acetic acid react to form nitrous acid, and nitrite reacts with p-aminobenzenesulfonic acid in the reagent to form diazobenzenesulfonic acid. Combined with α-naphthylamine to form a red product; bacteria such as Pseudomonas and Serratia can reduce nitrate or nitrite to nitrogen or nitrogen oxide.
This effect is called denitrification or denitrification, which produces bubbles but no red.

Operation steps (for reference only):
1. The tested bacteria are inoculated into a nitrate medium and cultured at 35 ° C for 1 to 4 days.
2. The appropriate amount of p-aminobenzenesulfonic acid solution and α-naphthylamine solution are mixed in equal amounts to be the nitrate reduction experimental reagent.
3. Take 0.1 ml of nitrate reducing experimental reagent, add to the medium, and observe the result immediately.
4. If the experiment is negative, add a little zinc powder to the test tube and observe the results immediately.
Dyeing results:

Note: There is no color reaction when adding reagents. Possible reasons: 1. Nitrate is not reduced, the experiment is negative; 2. Nitrate is reduced to other substances such as ammonia and nitrogen, causing false negatives. At this time, a little zinc powder can be added. If it is red, the experiment is negative. If it is not red, the experiment is false negative.
Precautions:
1. If you want to check if there is nitrogen, you can add a small inverted tube in the culture tube to see if there is any bubble.
2. Take self-protection in bacterial culture and experiment.
3. The culture time of the bacteria to be tested will also affect the dyeing. The culture time of the positive bacteria is too long, or the bacteria are dissolved, and the bacteria are often dissolved.
